A researcher has been given two CD4 T-cell clones from a collaborator. T-cell clone A is thought to be a Th17 clone specific for cytochrome C, and T-cell clone B is thought to be a Treg clone specific for myoglobin.

Question 1: Describe how you would design an assay to confirm that T-cell clone A indeed is a Th17 clone, and not a Th1 or Th2 clone.

Question 2: Describe how you would design an assay to establish whether T-cell clone B has suppressive Treg-cell function. Please justify your answer.
