You should be able to describe all procedures done in lab, know the use of all of the equipment, and all of the reagents used. You should be able to name at least one organism that is positive and one that is negative for each test we performed in lab.

1. Glo Germ Hand Wash System - Why do we wash hands? Describe proper hand washing technique.

2. Germicides (revised procedure) - What is the purpose of this test? How is this test performed? You should be able to interpret the results by observing incubated plates. You should be able to determine if an organism is resistant or susceptible to the disinfectants.

3. Methyl Red and Voges-Prosakuer Tests -What is the common substrate in these tests? What reagents are added to complete each test? What is the theory of each test? What does a positive result look like? What does a negative result look like?

4. Catalase Test - What does this test detect? What enzymes are involved? You should be able to describe the theory. What reagent is used? How is the test performed? What does a positive result look like? What does a negative result look like? You should be able to name at least one organism that gives a positive/negative result.

5. Oxidase - What does this test detect? What enzyme(s) are involved? Give the principle of the test. What type of respiration is involved? Name a genus of bacteria that yields a positive or negative test result. You should be able to distinguish a positive from a negative test result.

6. Nitrate Reduction - Give the principle of the test. Give the chemical reaction of Nitrate reducers. What enzyme(s) are involved? What are the possible end products? What reagents are added? What is the purpose of the zinc? You should be able to give the end products in the various tubes based on observations of the tubes. Name an organism that gives each result.

7. Citrate Test - Give the principle of the test. What is the substrate involved? What does growth indicate? What does a positive result look like? What does a negative result look like?

8. DNA Hydrolysis - What is the substrate? What is the enzyme? Describe the principle of the test. You should be able to distinguish a positive from a negative test result.

9. Urease Test- What is the substrate? What is the enzyme? What is the pH indicator? What is the end product? Describe the principle of the test. What genera is this test especially helpful in identifying? You should be able to distinguish a positive from a negative test result.

10. Bile Esculin Test - Give the principle of the test. What is the selective agent and what does it do? What is the indicator in the medium? What does a positive result look like? What does a negative result look like? Give an example of a positive organism.

11. SIM Test - Describe both pathways for the production of H2S. What is the substrate for hydrogen sulfide production in SIM media? What is the indicator? What other media did we use in class to detect H2S production? What other test results can be determined from the SIM tube? You should be able to interpret the results of each test.

12. Bacitractin - What group of Strep is this test specifically designed for? Discuss the theory of this test. How do you perform this test? You should be able to interpret the results from an incubated plate.

13. Novobiocin - What is the purpose of this test? You should be able to describe the theory of this test. How is this test performed? You should be able to interpret the results of this test.

14. Blood Agar - What kind of media is Blood Agar? What is hemolysis? Describe and tell what is happening in the media for each type of hemolysis. You should be able to interpret the results by observing incubated plates. Give one organism that demonstrates each type of hemolysis.

15. CAMP test - What group of Strep is this test specifically designed for? Discuss the theory of this test. How do you perform this test? You should be able to interpret the results from an incubated plate. What is the purpose of using the Staphylococcus aureus in this test?

16. Coagulase Test - What does this media test for? What enzymes are responsible? How is this test performed? You should be able to interpret the results by observing the incubated tubes. You should be able to name at least one organism that gives a positive/negative result.

17. Urine Culture - What is the purpose of this test? How is this test performed? You should be able to count colonies and calculate the number of organism per mL of the sample. You should be able to determine if the sample is an example of bacteriuria.

18. Kirby-Bauer Antibiotic Sensitivity Test - What is the purpose of this test? How is this test performed? You should be able to interpret the results by observing incubated plates. You should be able to determine if an organism is resistant or susceptible to the test antibiotics.
