1. Recognize the parts of the microscope and the function of each.

2. Recognize bacterial colony morphology, such as, colony shape, colony margin, colony elevation, pigmentation, and texture.

3. Recognize growth patterns in broth such as uniform fine turbidity, flocculent, sediment and pellicle.

4. Recognize the bacterial shapes: cocci, bacilli, and spirilla.

5. Recognize bacterial cell arrangement: diplo, strepto, staphylo, sarcina, and tetrad.

6. Be able to distinguish between a Gram stain, simple stain, endospore stain, capsule stain, acid-fast stain, negative stain and flagella stain.

7. Recognize a successful streaking for isolated colonies on a streak plate.

8 Be able to distinguish between lactose fermenters (Enterobacteriacea) and lactose non-fermenters that have been streaked on EMB agar and indicate the basis for the test.

9. Be able to distinguish between lactose fermenters (Enterobacteriacea) and lactose non-fermenters that have been streaked on MacConkey Agar and indicate the basis for the test.

10. Differentiate between alpha, beta and gamma hemolysis on a blood agar streak plate.

11. Differentiate between a positive and negative oxidase test.

12. Differentiate between a positive and negative catalase test and indicate the molecule responsible for a positive catalase test.

13. Differentiate between a positive and negative coagulase test and indicate the importance of this test.

14 Identify a positive gelatin liquefaction test and indicate the enzyme necessary for a positive test.

15. Identify a positive test for hydrolysis of milk protein and indicate the enzyme necessary for a positive test.

16. Recognize the production of gas, acid, or alkaline in a phenol red fermentation broth and identify the pH indicator associated with this test.

17 Identify a positive or negative hydrolysis of starch agar, if iodine has been added to the plate. Identify the enzyme that hydrolyses starch.

18. Identify a bacterium that is motile or non-motile when inoculated in motility agar.

19. Identify whether or not an organism is able to reduce nitrate to nitrite or some other nitrogenous compound, when inoculated in a nitrate reduction broth.

20. Identify whether or not an organism is able to reduce sulfur compounds when inoculated into SIM agar.

21. Indicate the presence or absence of a zone of inhibition with an antibiotic impregnated paper disc and indicate if the organism is susceptible, intermediate or resistant to the antibiotic.

22. Indicate the presence or absence of antigen in an agglutination reaction with blood types.

23. Indicate the use of micropippettors, how to set them for a specific volume.

24. Indicate function of SDS, proteinase K, and cold ethanol in extracting DNA from bacteria.

25. Distinguish among the following protozoa: Amoeba, Paramecium and Euglena, if given a wet mount or prepared slide of each.

26. Differentiate between yeast and molds, if given a microscopic or macroscopic view of either.

27. Differentiate between a positive and negative urease test and indicate the basis for the test.
