If I want it to work on the dark spot, how long do I have to leave it on before creaming it off (with a moisturizer) and applying makeup?

Also, the prescription information said DO NOT RUB IN, while an online source said to rub it completely in. I am putting this on with a toothpick so that I touch only the dark area and not the area around it. Rubbing it in would move the cream onto my normal skin.
